No, they are cousins. Check at the end of the episode when Conan has a flash back to the library mystery they say that Kaito's dad and Shinichi's dad are brothers. Therefor they are cousins.

I saw an episode or OVA not quite sure, where we see Kaitou's father talking with Shinichi's mother, and little cute Kaitou offers a rose to shinichi's mom!

Kaito and Shinichi aren’t brothers. As far as I recall, Gosho originally designed them to look alike.

I also don’t recall it being mentioned that Yusaku and Toichi were brothers. Actually, if I recall right, I think what was mentioned was that they “dueled” like Shinichi and Kaito. Also, Yukiko originally trained under Toichi, which would probably explain the rose from young Kaito.
