I dont know #1.
#2 is a negative. Example: Baka = stupid. Baka ja nai = not stupid. Simple as that. Used mostly with na adjectives, as well as verbs in some types of informal speech.

Vermouth just doesn't really lik it much i guess XD its fine to use tho' "ore, chisai koro kara kinina tten no ga irunsuyo!" = "I alraedy like someone and we knew each other from when we were little!" [Vol. 18 F.5 p18] "Arigatona, Chiisana Tantei-san." = "Thank you, Little Detective" [Vol.7 F.7 p18] Its all depending on where you us it I guess...^^ Chiisana is an adjective the first one is ... diffrent >>
